Mazdutide and survodutide activate two receptors, GLP-1 and glucagon. Retatrutide activates those two plus a third, GIP. All three belong to the newer group of incretin peptides that add glucagon receptor activity to the GLP-1 signal semaglutide is known for.

Why add glucagon to GLP-1

Glucagon and GLP-1 pull blood glucose in opposite directions. In research models, glucagon receptor activity raises energy expenditure and fat oxidation in the liver, but on its own it also raises glucose. GLP-1 receptor activity lowers glucose. A peptide that does both is designed to keep the energy-expenditure side of glucagon while GLP-1 offsets its effect on glucose.

That design goal is the reason all three of these molecules exist. Where they differ is in how the two (or three) receptor signals are balanced, which each developer tuned separately.

Mazdutide

Mazdutide is modeled on oxyntomodulin, a natural gut hormone that already activates both the GLP-1 and glucagon receptors. It carries two development codes: LY3305677 from Eli Lilly, where it started, and IBI362 from Innovent Biologics, which developed it under licence. It is studied in metabolic and energy-balance research. We sell mazdutide in a 10 mg vial.

Survodutide

Survodutide, development code BI 456906, is a GLP-1 and glucagon dual agonist from Boehringer Ingelheim, developed with Zealand Pharma. Alongside metabolic research, it has been studied in liver-fat models, which fits the hepatic side of glucagon receptor activity. We sell survodutide in a 10 mg vial.

Retatrutide

Retatrutide, Eli Lilly's LY3437943, is the triple agonist of the three: GLP-1, GIP and glucagon. Its molecular weight is 4731.32 g/mol (formula C221H342N46O68, CAS 2381089-83-2). Identifiers we publish, and the ones we do not yet

Retatrutide's formula, mass and CAS number are printed on its product page. For mazdutide and survodutide we have not printed a formula or CAS number yet. Published reference values for newer peptides are sometimes inconsistent, and we would rather print nothing than print a wrong number that looks authoritative. Every vial's own lab report still states the observed mass for its batch, and the QR code on the vial opens that report.

Where the amylin peptides fit
